Biotin (vitamin B8 or H) at 2.5mg (2500µg) is a pharmacological dose significantly above the NRV (50-150µg/day depending on the country). The 2.5mg dose is commonly used for beauty goals (hair, skin, nails) and for diagnosed deficiencies. Biotin is an essential cofactor of carboxylases (acetyl-CoA carboxylase, pyruvate carboxylase), key enzymes in carbohydrate, lipid, and protein metabolism. For athletes, biotin supports fatty acid synthesis (membrane recovery), gluconeogenesis (maintaining blood sugar during endurance), and branched-chain amino acid synthesis. The 2.5mg dose is considered safe because biotin is water-soluble with rapid renal elimination (no toxic accumulation).
Is Biotin 2.5mg genuinely effective for improving hair and nail quality, or is it mainly a marketing myth?
The answer is nuanced. Biotin deficiency does indeed cause alopecia (hair loss) and brittle nails. Supplementation correcting this deficit restores hair and nail quality. For people WITHOUT biotin deficiency (the vast majority of the population with a normal diet), clinical evidence of hair improvement from biotin is very limited. Zempleni et al. and other researchers point out that positive studies generally included deficient individuals. Biotin needs are increased in certain specific situations: pregnancy and breastfeeding, regular consumption of raw egg whites (avidin in raw egg white binds biotin and prevents its absorption), prolonged antibiotic treatment (impairs the biotin-producing gut flora), high alcohol consumption. For athletes without these risk factors, biotin needs are not significantly increased by training. Athletes eating whole cooked eggs (cooked egg white no longer has anti-biotin properties) in their diet generally cover their needs. Direct evidence of athletic performance improvement from biotin is nonexistent in the scientific literature. Biotin acts as a basic metabolic cofactor (energy metabolism), but its supplementation does not generate measurable ergogenic effects in people with normal levels. It is not a sports ergogenic aid in the classic sense (no direct gain in strength, power, endurance, or body composition). Its value for athletes is therefore mainly indirect: support for general energy metabolism (particularly carbohydrate metabolism), skin integrity for athletes with intense sweating, and correction of any deficiencies. The cost-benefit ratio of Biotin 2.5mg for pure athletic performance is low.
Can Biotin 2.5mg be taken daily without risk, and does its renal elimination prevent any risk of toxic accumulation?
Biotin is a water-soluble vitamin with rapid renal elimination: excess is excreted in urine, with no known risk of toxic accumulation even at very high doses (up to 10mg/day in clinical trials on multiple sclerosis with no observed toxicity). EFSA has not established an upper safety limit for biotin due to the absence of documented toxicity. Biotin 2.5mg can therefore be taken daily safely from a toxicological standpoint. The only known clinical risk is interference with biological tests (see previous FAQ) — always inform your doctor. For people with impaired kidney function (renal insufficiency), reduced renal elimination may increase blood concentrations; consult your nephrologist.
